The three DNA fragments were concatenated into one DNA construct by overlap extension PCR (Horton et al., 1993) for transformation. As a control, a transformant with the pyrG marker downstream of native ustA was prepared in ustROE pyrG- by using a DNA construct composed of ∼ 1-kb ustA upstream, ustA, pyrG, and ∼ 1-kb ustA downstream (Fig. S3).
