Post-translational modifications inducing proteasomal degradation to counter HIV-1 infection
2020, Virus ResearchCitation Excerpt :Likewise, multiple viral proteins are involved in degradation of the identical cellular target molecules, and thus specific contributions from each viral protein to virus replication via proteasomal degradation of the target protein may be nearly immeasurable. For example, both Env and Nef affect SERINC-associated virus entry (Chande et al., 2016; Inuzuka et al., 2005; Rosa et al., 2015; Shi et al., 2018; Usami et al., 2015; Wu et al., 2019), Vpu/Env/Nef regulate down-modulation of CD4 on the HIV-1-infected cell surface (Estrabaud et al., 2007; Garcia and Miller, 1991, 1992; Geleziunas et al., 1991; Margottin et al., 1998; Petersen et al., 1992; Schaefer et al., 2008), and so on. However, the relative influence of each viral protein, such as Vpu/Env/Nef, on CD4 reduction is elusive.
Efficacy and safety of mycophenolate mofetil versus levamisole in frequently relapsing nephrotic syndrome: an open-label randomized controlled trial
2019, Kidney InternationalCitation Excerpt :Peripheral blood mononuclear cells were incubated at 2 × 106 cells/ml per well, with phorbol myristate acetate (50 ng/ml), ionomycin (500 ng/ml), and monensin (2 μmol/ml) for 4 hours at 37 °C in 5% CO2.51 Surface staining with anti-CD3 and anti-CD8 antibodies was followed by fixation, permeabilization, and incubation with anti–interferon-γ, anti–interleukin-13, and anti–interleukin-17 to identify Th1, Th2, and Th17 cells, respectively, on T-helper cells (CD3+, CD8-), using fluorescence –1 to gate the populations.52 Cells were acquired on a BD LSR II flow cytometer and data were analyzed with BD FACS Diva version 6.0 (Becton, Dickinson, San Jose, CA).
Beneficial therapeutic effect of Chinese herbal Bushen formula on CHB patients with mildly elevated alanine aminotransferase by down-regulating CD4+CD25+T cells
2013, Journal of EthnopharmacologyCitation Excerpt :The antiviral activity of IFN-γ against HBV replication has been demonstrated in HBV transgenic and hepatocyte cell lines (Guidotti and Chisari, 2001; Robek et al., 2002; Park et al., 2005; Parvez et al., 2006). Petersen CM et al. reported that phorbol esters down-regulated the cellular CD4 pool by endocytosis and subsequent lysosomal degradation of membrane CD4 (Petersen et al., 1992). Therefore, in this study, CD3+CD8− cells were considered as CD4+T cells.
