Bronchoprovocation Testing in Asthma: An Update
2018, Immunology and Allergy Clinics of North AmericaCitation Excerpt :Adenosine exerts its effects through interaction with specific adenosine (P1) receptors. The exact mechanism of action of AMP on airway wall is not well-defined, but the role of mast cells is crucial based on a variety of inflammatory mediators, such as histamine, prostaglandins, leukotrienes, and IL-8 release, from human mast cells enhanced by adenosine in vitro.156–159 Although there are studies that have shown differential BHR response to antiinflammatories such as ICSs, there are also those that show concordant effects of these medications on bronchial responsiveness to direct and indirect stimuli.143,160–163
Adenosine activates Gα<inf>s</inf> proteins and inhibits C3a-induced activation of human mast cells
2018, Biochemical PharmacologyCitation Excerpt :Adenosine alone stimulates the production of interleukin (IL)-4 and IL-8 in the human MC line HMC-1, which expresses A2A and A2B receptors [31]. In clear contrast, adenosine inhibits IgE-induced degranulation of human skin MC [32] and dispersed human lung MC [33] through Gαs-coupled adenosine receptors. Similar to A1 and A3 receptors, P2Y receptors are Gαi-coupled purinergic receptors [34].
