The use of a very sensitive nested PCR method allowing the detection of subclinical infections by P. jirovecii has been described (Armbruster et al., 1997; Leigh et al., 1993; Nevez et al., 1999; Olsson et al., 1993). In concurrence with these reports, we could speculate that the patient classified in group 3 (possible PcP), who had a positive mtLSUrRNA-PCR, probably suffers from a subclinical P. jirovecii infection.

Single-step PCR and Southern blot hybridization have been successfully used to detect P. jirovecii in bronchoalveolar lavage (BAL) and induced sputum samples (Leigh et al., 1993; Lipschik et al., 1992). PCR, nevertheless, requires improvements to become a routine diagnostic test for PCP.
